miRNA expression profile of retinal pigment epithelial cells under oxidative stress conditions: a study from the Antonella Sidoti’s group.

Deep analysis of regulative mechanisms of transcription and translation in eukaryotes could improve knowledge of many genetic pathologies such as Retinitis pigmentosa (RP). New layers of complexity are daily discovered, such as the preponderant role in regulative functions of miRNAs. We compared changes in the expression of miRNAs obtained from whole transcriptome analyses, between two groups of retinal pigment epithelium (RPE) cells, one untreated and the other exposed to the oxidant agent oxidized low-density lipoprotein (oxLDL), examining four time points (1, 2, 4 and 6 h). We found that 23 miRNAs exhibited altered expression in the treated samples, targeting genes involved in several biochemical pathways, many of them associated to RP for the first time, such as those mediated by insulin receptor signaling and son of sevenless. Moreover, five RP causative genes (KLHL7, RDH11, CERKL, AIPL1 and USH1G) emerged as already validated targets of five altered miRNAs (hsa-miR-1307, hsa-miR-3064, hsa-miR-4709, hsa-miR-3615 and hsa-miR-637), suggesting a tight connection between induced oxidative stress and RP development and progression. This miRNA expression analysis has discovered new regulative functions of miRNAs in RP that should lead to the discovery of new ways to regulate the etiopathogenesis of RP. Researchers of the Italian National Research Council (CNR) and of the Euro-Mediterranean Institute of Science and Technology (I.E.ME.S.T.) have published a new research on the “Journal of Proteome Research”, about the early mechanisms of Alzheimer’s disease (AD). In particular, a neuronal cell model was treated with the Aβ peptide, one of the principal agent in AD, to understand which biological mechanisms are modulated in the early stage of the disease. Dr Luigi Inguglia, a researcher affiliated to the I.E.ME.S.T., has used a bioinformatic approach to analyze the mass spectrometry data, from the treated and untreated cells, describing the down-regulation of important factors of the ribosome biogenesis and of the splicing processes. These data, confirmed by immunoblotting assays, show that the dysregulation of the RNA metabolism mechanisms and of the spliceosome components represent a key early event in the Alzheimer’s disease.

An international team of scientists including University of Palermo and IEMEST researchers and colleagues from Poland (Medical University of Gdansk) and U.S.A. (University of Maryland) demonstrated a new action mechanism of the drug doxorubicin in vitro. This mechanism involves the induction of replicative senescence through treatment with low doxorubicin doses inducing HSP60 acetylation. In the lung cancer cell line studied, Hsp60 forms a complex with the onco-suppressor protein p53 favoring tumor progression. Hsp60 acetylation interferes with the formation of the Hsp60/p53 complex promoting its dissociation, causing an increase in the levels of free p53, which can then activate the p53-dependent pathway toward cell senescence. The work was published this month in the journal Cancer Letters (Impact Factor: 5,992) and can be downloaded from the site http://www.sciencedirect.com/science/article/pii/S0304383516306760

Research anticancer: Palermo researchers involved in the discovery of a chemotherapy that blocks the cancer cells.
In a study recently published in the international journal "Epigenetics", researchers from the Euro-Mediterranean Science and Technology (Manlio Vinciguerra, Francesca Rappa and Francesco Cappello), in collaboration with Italian English, Spanish, Estonian, Czech and US scientists, have demonstrated the effects and efficacy of guadecitabine, a chemotherapic agent, useful in preventing the progression of HCC, both in vitro through the use of cell models and in vivo through the use of murine models. The study was coordinated by Manlio Vinciguerra while Francesca Rappa and Francesco Cappello conducted experiments and morphological evaluations.

Hepatocellular carcinoma is linked to epigenetic alterations such as overexpression of variants of histone proteins and hypermethylation of the promoters of some tumor suppressor genes: all these alterations are currently target by the research for new chemotherapeutic drugs.

The chemotherapeutic drug studied in this scientific work is a second generation hypomethylating agent and is able to block the growth of cancer cells.

One of the most interesting aspects of the work is that the guadecitabine has also shown effective on the hepatocellular carcinoma cell models that have an overexpression of the histone variant macroH2A and are resistant to first-generation hypomethylating agents.

A new mechanism to fight liver cancer showed by IEMEST’s researchers.
An international team of scientists including IEMEST researchers and colleagues from U.K. (University College of London, Queen Mary University of London e Nottingham Trent University), The Netherlands (University Medical Center of Groningen), Estonia (Tallin University of Technology), Australia (University of Sidney) and Spain (Campus Can Ruti), showed for the first time a mechanism by which the hepatocellular carcinoma, one of the most aggressive liver cancer, is able to resist to chemotherapy. This mechanism involves the histone protein macroH2A1. The latter is now proposed as a novel target for the therapy of this tumor - very frequent in the Western Countries – which onset is associated to some infections, alcohol abuse, metabolic diseases and pollutants. Our group, formed by Francesca Rappa, Francesco Cappello and Manlio Vinciguerra (the latter is the Corresponding Author of the paper) performed a number of experiments including those to establish, morphologically, the hepatic damages, the subcellular localization of the histone proteins and their levels in the different experimental models that have been used in this study. Cancer Research (I.F.: 9,329) is the most frequently cited cancer journal in the world. A NEW PUBLICATION FROM IEMEST'S RESEARCHERS EXPLAINS ONE OF THE MECHANISMS OF ACTION OF A CHEMOTHERAPICS
A group of IEMEST researchers, lead by Dr. Claudia Campanella, Head of the Section of studies on micro- and nano-vesicles as biomarkers, has showed for the first time one the mechnisms of action of a known chemotherapics, the SAHA (suberoylanilide hydroxamic acid). This mechanism involves the nitration of the mitochondrial protein HSP60. The latter, after nitration, is released from tumor cells by exosomes. HSP60 commonly protects tumor cells from stress. Its secretion through exosomes favours tumor cell death as well as activates an antitumor immune response. On November 13th and 14th, during the conference “Science in Ancient / Science for Ancient. From scientific knowledge in the Hellenistic age to the virtual archeology” organized by the “Regione Siciliana”, the IEMEST researcher Francesco Di Paola will give a Lecture on “The Morgantina Silver Archaeological Treasure at the Museum of Aidone” with other colleagues in the field of Computer Science. The Morgantina Treasure is one of the most precious collections of the Archaeological Museum in Aidone, consisting of 16 pieces that were returned to Italy in 2010, following the 2006 agreement between the Italian and Sicilian governments and the Metropolitan Museum in New York. A police investigation determined that the provenance of the objects was the house of Eupolemos in Morgantina, where the precious items probably were hidden during the Second Punic War. Before the trip to New York the research team have been decided to examine the silver pieces through non-invasive diagnostic analysis and 3D survey to collect useful data both to monitor the conservation state over time (to check after four years) and to guarantee the virtual visit of the item during their absence.

A Potential Link between lipoproteins and asthma: a IEMEST's study
A recent study carried out by two research groups coordinated by Prof. Scichilone and Prof. Rizzo, IEMEST researchers, demonstrated for the first time that, in asthmatics, the least pro-inflammatory lipoproteins (LDL-1 e LDL-2) are reduced, and the pro-inflammatory (LDL-3 and LDL-4) elevated compared to healthy controls. In addition, in asthmatics the serum concentrations of LDL-1 are positively associated with lung function, whereas the pro-inflammatory LDL-3 are inversely correlated with lung function. These findings suggest that the LDL are implicated in the pathogenesis of asthma, in that, they may act by igniting the inflammatory cascade of the airways.

A new study correlates obesity associated with insulin resistance to Alzheimer's disease
Two researchers of our Institute are co-authors of a study carried out along with researchers of the C.N.R. of Palermo, which correlates the insulin resistance, that develops in some obese individuals, with the development of Alzheimer's disease. The study has been published on "Current Alzheimer Research". In particular, insulin resistance determines, among other damages, oxidative stress and mitochondrial dysfunction in neurons of the mice subjected to a high fat diet, and this leads to early damages that are pathognomonic for Alzheimer's disease. Treatment of metabolic disorders, then, can prevent the development of this serious neurodegenerative disease in this subpopulation of subjects. Part of this work was carried out by using the equipment of the Laboratory of Neuroanatomy funded by PON 'Research and Competitiveness' 2007-2013, measure 4.1.1.4.

A novel biomarker for the colo-rectal cancer
A group of IEMEST’s researchers, coordinated by prof. Francesco Cappello, published a paper in the journal "CANCER" (the official journal of the American Cancer Society) on a new marker for the diagnosis and the follow-up of patients with colorectal carcinoma. In this work it has been shown that a stress protein (Hsp60) bound to exosomes (vesicles secreted by tumor cells) is present in very high levels in patients who undergo surgery while its levels are reduced drastically one week after the surgery. These results make exosomal Hsp60 a putative tool for the diagnosis and the follow-up of this cancer. The work was conducted in collaboration with researchers of the University of Palermo, the National Institute of Health in Rome and the University of Maryland at Baltimore, USA.

PROGETTO CYBER BRAIN: PRIMA PUBBLICAZIONE SU RIVISTA “NATURE”

The results of a study carried out by the Department of Medicine and Neuroscience of the Euro-Mediterranean Institute of Science and Technology (IEMEST) has been published on March 24th in an international journal, part of the Nature publishing group, called "Scientific Reports". Dr. Antonella Marino Gammazza, the principal investigator of the study, said: “The study is among the first to explore the relationship between temporal lobe epilepsy (TLE), the most frequent type of non-treatable epilepsy, and the expression of one of the most important anti-stress proteins, the Hsp60. The work was conducted using both an animal model and human samples. In the animal model, the level of Hsp60 was evaluated in the hippocampus and in blood. An increase of Hsp60 levels was observed not only in the hippocampus, but also in the plasma of the animals following epileptic seizures. The correlation between the increased expression of the protein in circulation and the high neuronal excitability is very interesting. Likewise, post-seizure Hsp60 levels in the plasma of patients were higher than before the seizure. In conclusion, high levels of Hsp60 could be considered a biomarker of stress, reflecting neuronal cell damage in the hippocampus”.

This study was conducted in collaboration with researchers from the IRCCS "NEUROMED" (Isernia, Italy), and is the first study conducted using the equipment acquired through the "CYBER BRAIN" PON 2007-2013 project, funded by the Ministry of Education (Project Code: PONa3_00210), for the establishment of an advanced neuroanatomy laboratory in Palermo at the IEMEST. Researchers from the University of Maryland, Baltimore, USA, from the University of Cardiff, UK, and from the University of Malta also took part in the work. The President of the IEMEST, Bartolomeo Sammartino, and the Director, Francesco Cappello, would like to express their satisfaction for the important goal achieved thanks to the PON funding received by the Institute and to the collaboration with other partners, including the IRCCS "Neuromed". A researcher at the Institute for Euro-Mediterranean Science and Technology in the Top Italian Scientist.

Prof. Giovanni Li Volti, Director of the Department of Stress Biology, Epigenetics, Sport and Health has entered the ranks of the Top Italian Scientist elaborated by VIA-Academy. This ranking includes Italian scientists of all disciplines (science and humanities) operating in Italy and in the world. The ranking is compiled based on h-index, the index that measures the number of international publications, and their impact on the international scientific community assessed on the number of citations received. Among the names that stand out in the top of the rankings we found researchers such as: Carlo Croce, Giuseppe Remuzzi, Umberto Veronesi, Silvio Garattini, Carlo Rubbia and Antonio Zichichi. "I feel very honored to be part of such a prestigious ranking," said Prof. Li Volti "I feel proud to represent my country and particularly young researchers of the south of our country." "Obviously," says Li Volti, "we must take into account the limitations of the standings. The h-factor functions primarily as a comparison among researchers of the same subject area, and favors those who have a long career behind compared to the young, as brilliant. The index varies in fact considerably between different fields (for example, is quite high in the biomedical field and in physics). Furthermore, use of these bibliometric data usually disadvantages researchers not publishing in English"

An important study about aging and liver diseases

The process of aging predisposes to hepatic functional impairment. Therefore, understanding how aging could affect the molecular pathology of liver diseases is important. In the review “Liver disease and aging: friends or foes?” appeared on the prestigious journal Aging Cell, our IEMEST researcher Dr. Manlio Vinciguerra, also Senior Lecturer at the University College London, discusses why common liver diseases such as non-alcoholic fatty liver disease, non-alcoholic steatohepatitis, cirrhosis and hepatocellular carcinoma surprisingly occur less frequently in the very elderly (individuals aged more than 70).

CHRONIC OBSTRUCTIVE PULMONARY DISEASE:

NEW EVIDENCES ON ITS PATHOGENESIS

Published in PLOS ONE

Chronic obstructive pulmonary disease (COPD) is one of the most severe conditions caused primarily by cigarette smoke and characterized by a progressive airway obstruction. Many people each year die from complications of this disease worldwide. In a study conducted by researchers from many institutions - among which the IEMEST, the "S. Maugeri " Foundation, and the Universities of Palermo, Ferrara, Torino and Padova, new molecules involved in induction and maintenance of the inflammation that underlies the obstruction of the airways have been identified. <<These molecules are called "chaperones" or "heat shock proteins">> says prof. Cappello, principal investigator in this study, <<and while they are physiologically devoted to protect cells from stress induced by various factors (e.g., cigarette smoking) in this disease some of them are overexpressed and they may increase the activity of inflammatory cells . In this paper we describe for the first time some molecular mechanisms involved in its overexpression. These results may lead to more effective therapies for COPD>>.

Cordonal stem cells as new promising therapeutic tools

Cells from the Umbilical cord’s Wharton jelly (WJC) are promising tools for the treatment of cancer, muscolo-skeletal, neurodegenerative, hepato-pancreatic diseases. Following the recent advances in the field, the international scientific journal “The Open Tissue Engineering and Regenerative Medicine Journal” just published a special issue titled “Wharton’s jelly mesenchymal stem cells: tissue regeneration and beyond”, guest edited by Dr Giampiero La Rocca, IEMEST researcher. Contributions were from many pioneers in the field of WJC: International authors were from top laboratories in Belgium, Canada, Germany, India, Japan, Switzerland and USA. Italian groups were from the Universities of Chieti, Milano, Padova and Palermo. Different aspects of WJC biology, immune features, disease indications, were discussed in a series of authoritative reviews. Selected original papers provide novel findings on isolation, culturing, features and uses of WJC for cellular therapy. Taken together, the issue provides key advances on the definition of the new frontiers which can be reached, further moving on the current targets of WJC-based regenerative medicine.

New discovery: two new molecules predictive for acute myocardial infarct

In this pilot study published in Translational Research (2011;157:285–292), we determined serum levels of Hsp60 and Hsp32 in 40 patients with acute myocardial infarction (AMI) to assess their clinical significance and potential prognostic value. We also performed a bioinformatics analysis of the two molecules in search of structural clues on the mechanism of their release from cells. All patients were followed up for 6 months to register adverse post-AMI cardiovascular events

A multivariate analysis demonstrated that elevated Hsp60 was predictive of post-AMI adverse events. In contrast, increased Hsp32 levels showed a significant association with less severity of coronary artery diseases. Moreover, our findings suggest that Hsp60 and Hsp32 play distinct roles in the pathogenesis of AMI.

A new discovery on inflammatory bowel diseases published on “Cell Stress and Chaperones”

The purpose of this work was to determine in colon mucosa of Crohn’s disease (CD) and ulcerative colitis (UC) in relapse: a) the levels of the chaperonins Hsp60 and Hsp10; b) the quantity of inflammatory cells. Both Hsp60 and Hsp10, as well as inflammatory cells, occurred in the mucosa of CD and UC but not in NC. Hsp60 and Hsp10 co-localised to epithelial cells of mucosal glands but not always in connective tissue cells of lamina propria, where only Hsp60 or, less often, Hsp10 was found. Since chaperonins are key factors in the activation of the immune system leading to inflammation, we propose that they play a central role in the pathogenesis of the two diseases, which, consequently, ought to be studied as chaperonopathies.

New discovery:

A protein secreted by tumor cells may become a new diagnostic and therapeutic target

published in PLoS ONE

Levels of Hsp60 in tumors are often at high levels. Moreover, Hsp60 may be found in sera of patients with tumor. Data presented in this work suggest that Hsp60 release from tumor cells is the result of an active secretion mechanism and, since extracellular release of the chaperone was demonstrated in all tumor cell lines investigated, our observations most likely reflect a general physiological phenomenon, occurring in many tumors. Finally, we showed that Hsp60 is present in tumor cell membrane. These results may have diagnostic and prognostic, as well as therapeutic significance.

New discovery: A new scaffold to differentiate cardiac precursors in myocardiocytes

Published in Journal of Cellular Physiology

In the last few years, a major goal of cardiac research has been to drive stem cell differentiation to replace damaged myocardium and several research groups have attempted to differentiate potential cardiac stem cells (CSCs). In the present study, we isolated a pool of small proliferating and fibroblast-like cells from adult rat myocardium. The phenotype of these cells was assessed and the characterized cells were cultured in a collagen I/OPLA scaffold with horse serum to obtain fine myocardial differentiation. The results open new biomedical and therapeutical scenarios.

New discovery: Chlamydia Trachomatis infection consequences: autoimmune diseases or anticancer immunity?

published in PLoS Pathogens

Chlamydia trachomatis (CT) infection is one of the most common causes of reproductive tract diseases and infertility. This article focuses on the potential pathogenetic effect of Hsp60 production from Chlamydia trachomatis that shares various antigenic determinants with the human counterpart. At the same time, we remark that pre-tumor and tumor cells expressing Hsp60 on their surface can be destroyed with participation of the anti-Hsp60 antibody, thus stopping cancer progression before it is even noticed by the patient or physician.
